醋酸纖維素經溶劑溶解後再塗佈揮發而成之薄膜,能符合製備鑲嵌式生物感測器之超薄、強勒、有效固定酵素等要
求。本實驗將葡萄糖氧化?固定於醋酸纖維素膜,再和溶氧計測系統組合成葡萄糖感測器。該感測器於批式操作時,測
定一個樣品需時2.5分鐘;葡萄糖濃度範圍在400 ppm以下時,測得之溶氧降低值與葡萄糖濃度之間呈良好直線關係;常
見之醣類中除了葡萄糖以外,只對果糖和蔗糖有5%以下之反應;眾多金屬離子中,只有 Fe 和Hr 有顯著影響。此外
,該感測器對同一樣品有良好再現性,平均相對誤差僅在2%左右;並和常被用於吸光法定量還原糖之雙硝基水楊酸
(DNS)呈色法之間,有良好相關性,經四批樣品測試比對結果,相關係數均在0.99以上。
由於此一葡萄糖感測器計測系統具備優異之再現性及長期安定性,因而被認為頗能切合醱酵程序連續計測之需求,
實際應用之相關試驗及其結果,將於下一篇報告中詳細列述討論。
m - 英文摘要: A glucose determination system by incorporating immobilized glucose oxidase membrane in a dis-solved oxygen detection system was evaluated for use in continuous monitoring of the fermentation pro-cess. Cellulose acetate was selected as the insoluble carrier for the preparation of immobilized enzymemembrane. The procedure for constructing this system, including the optimization of membrane prepara-tion and enzyme immobilization, is described in detail. Factors influencing the response of the sensor sys-tem were also investigated. The response time and recovery time for this sensor were 90 and 60 seconds,respectively. More than 20 samples can be analyzed within an hour by the batch type operation and steadystate method. The stability and reproducibility of this sensor were satisfactory. And, there is a good cor-relation with the DNS method. Good stability and reproducibility favored this sensor system for applica-tion in continuous monitoring of the glucose level in the fermentation process.
