- 中文摘要: 以腫瘤胚胎抗原(CEA)致敏之小鼠脾臟細胞與小鼠骨髓瘤細胞株P3-NS1/1-Ago融合後得到4個融合瘤,產生4種屬IgGl之單源抗體,以抑制性放射免疫分析測知4種單源抗體對正常人大腸組織之過氯酸抽取物均無反應。單源抗體MAbC27對CEA之親和性最好。其與各種人大腸直腸腺癌細胞株作螢光染色反應之強度與癌細胞分泌CEA之能力是一致的。如LS174T及SW1083二種細胞株之陽性率最高(95%)而COL0320DM及COL0320HRS為最低(0.5%),以免疫酵素染色法染正常人之大腸、肺、肝、脾及腎臟等組織切片均無反應,而對多種癌組織切片染色之陽性率分別為:大腸直腸腺癌100%(30/30),胃癌96.6%(28/29),肝癌32.1%(9/28)及肺癌82.1%(69/84)以上結果顯示MAbC27似可作為大腸直腸腺癌之免疫偵測及同位素掃描定位。
- 英文摘要: Four hybridomas secreting monoclonal antibodies (MAbs) of the IgGl subclass against human car-cinoembryonic antigen (CEA) were obtained from fusion of P3-NS1∕1-Ago myeloma cells with splenic cellsfrom mice immunized with purified CEA. None of the MAbs showed cross-reactivity to perchloric acidextractable antigens from the normal human colon by an inhibition radio immunoassay. However, MAb C27showed the highest affinity to CEA. The intensity of immunofluorescence staining of human colorectalcancer cells with MAb C27 correlates well to the cellular CEA content of cancer cells. LS174T showed thehighest intensity of fluorescence (95%) while COL0320DM and COLO320HRS were the lowest (0.5%).None of the normal human organs-colon, lungs, liver, spleen or kidneys-showed positive staining byimmunoperoxidase anti-peroxidase (PAP) techniques, while tissues from colorectal carcinoma (CRC),gastric carcinoma, hepatoma and lung cancer gave a positive rate of 100% (30/30), 96.6% (28/29), 32.1%(9/28) and 82.1% (69/84) respectively. Results suggest that MAb C27 can be used in immunodetection andradiolocalization of colorectal carcinoma.
