- 中文摘要: 去勢動物若再切除甲狀腺,血中促黃體素(LH)濃度會增高。為探討其中原因,本報告以離體實驗,觀察大白鼠腦下腺前葉對性釋素(GnRH)之反應能力是否會因甲狀腺之切除而改變。成年大白鼠於去卵巢後分為三組,第一組立即切除甲狀腺,第二組於甲狀腺切除後補償甲狀腺素(T4)。第三組為假手術組。所有動物於45天後犧牲,取出腦下腺前葉,和各種劑量之性釋素培養4小時,每一劑量均為四重複。培養液及血清中之促黃體素及促甲狀腺素(TSH)以放射免疫法測定。結果發現在每毫升0.1 ng之性釋素刺激下,去甲狀腺鼠的腦下腺比假手術者及甲狀腺素補償者釋出較多的促黃體素。這些結果指出,去卵巢鼠於切除甲狀腺後,腦下腺前葉對性釋素反應能力的提高是導致血中促黃體素濃度增高的因子。
- 英文摘要: The purpose of this investigation was todetermine if thyroidectomy affects pituitary LHreleased from ovariectomized (Ovx) rats inresponse to GnRH in vitro. Adult rats wereboth Ovx and thyroidectomized (Tx) or shamTx. The Ovx-Tx rats were injected sc witheither saline or T4 (2μg/l00g BW/day) oncedaily for 45 days. Twenty-four hours after thelast injection, all rats were sacrificed by deca-pitation and the blood was collected from thebody. Anterior pituitaries (APs) were removedand incubated in vitro with synthetic GnRH at37℃ for 4 h. Two hemi-APs were placed inone flask and quadruplicate determination wasmade for each group. Concentration of LH inthe medium and serum was determined byradioimmunoassay. LH in the serum was 62%higher (p<0.01) in Tx rats than in sham Txrats. Following incubation with 0.1 ng GnRHthe LH released by the APs of Tx animals was84% (p<0.05) and 94% (p<0.05) higher thanthose of sham Tx and Tx-T4 rats, respectively.These results suggest that the stimulatory effectof thyroidectomy on serum LH in Ovx rats ismediated at least in part by the enhancementof the responsiveness of anterior pituitaries toGnRH.
