本研究為利用人類絨毛膜性腺激素為免疫劑,以製造抗人類絨毛膜性腺激素單源抗體。Balb/c小白鼠經過hCG與免
疫佐劑混合免疫注射四次後,選取抗體力價最高之鼠於細胞融合3天前再行靜脈注射免疫一次。犧牲此小白鼠後採其
脾臟細胞與骨髓瘤細胞NS-1,以4:1比例混合,以PEG 4000行融合反應,經過HAT培養液之篩選後,以尿素?或過氧化
?免疫反應及放射免疫分析法來測定其抗體之產生。得到能產生抗人類絨毛膜性腺激素之融合瘤之後再以極限稀釋法
進行分株兩次以建立單源細胞株,然後再交互以?免疫分析及放射免疫分析測定其上清液中之抗體產生情形。此細胞經
過大量培養後,取此細胞以之冷凍或以之注射至前以Pristane處理之小白鼠腹腔中以產生腹水,而取上清液作抗體之鑑定
。結果顯示以PEG當融合劑時,融合率在42.0至50.0%之間。總共產生$42個融合瘤。其中403個可以產生人類絨毛膜激
素抗體。經過極限稀釋法產生單源細胞株及再次以?免疫反應及放射免疫分析後,剩下39個融合瘤細胞株可以產生人類
絨毛膜性腺激素特異抗體。其中57-42-2融合瘤細胞株產生抗體能力最強。以之來作抗體之定性。以Outchterlony免疫
擴散法,可鑑定其免疫球蛋白類別為IgG。以SDS-PAGE及放射免疫沉澱分析,可見其對hCG具有免疫反應作用。此單源
抗體與hCG間之親和力常數以Scatchard plot法得到為0.6x109 1/mol。在放射免疫分析中此抗體與LH, FSH之交互反應
能力為1.5%及0.7%,與TSH則無交互反應之情形。c - 英文摘要: Production of monoclonal antibodies against human chorionic gonadotropin (hCG) has been studiedusing hCG as an immunogen. Spleen cells of BALB/c mice immunized with hCG were fused with NS-1mouse myeloma cells. This study reports the successful isolation of a hybrid clone secreting a monoclonalantibody specific for hCG. By using PEG 4,000 as a fusion agent, the fusion rates were between 42.0 and50.2%. In total 842 hybridomas were produced. Among them, 403 hybridomas had hCG antibody produc-tion. After cloning twice by limiting dilution and alternately screening by enzyme immunoassay and byradioimmunoasay, there were 39 cell lines having specific antibody production. Among them, the No.57-42-2 had the highest reactivity. By Ouchterlony test, the monoclonal antibody was shown to be IgG1.The affinity constant of the antibody to hCG was 0.6 x 109 1/mole. In radioimmunoassay, the crossreactivity of the antibody to human luteinizing hormone (LH) and human follicle-stimulating hormone(FSH) was 1.5% and 0.7%, respectively. There was no cross reaction with human thyrotropin-stimulatinghormone (TSH).
