- 英文摘要: Chromosomal instability(CIN)refers to high rates ofchromosomal gains and losses and is a major cause ofgenomic instability of cells. It is thought that CIN causedby loss of mitotic checkpoint contributes to carcinogene-sis. In this study, we evaluated the competence of mitoticcheckpoint in hepatoma cells and investigated the causeof mitotic checkpoint defects.We found that 6(54.5%)ofthe 11 hepatoma cell lines were defective in mitoticcheckpoint control as monitored by mitotic indices andflow-cytometric analysis after treatment with microtu-bule toxins. Interestingly, all 6 hepatoma cell lines withdefective mitotic checkpoint showed significant under-expression of mitotic arrest deficient 2(MAD2),a keymitotic checkpoint protein.The level of MAD2 underex-pression was significantly associated with defectivemitotic checkpoint response(p<0.001).In addition,nomutations were found in the coding sequences of MAD2in all 11 hepatoma cell lines.Our findings suggest thatMAD2 deficiency may cause a mitotic checkpoint defectin hepatoma cells.
