新鮮豬胰臟可以用丙酮乾燥法製備成粉末,對每一百克新鮮豬胰臟收量約為十至十五克。蛋

,可得較高之Carboxypeptidase A 和 B 之活性,由胰臟粉末之抽提液中活化時,則可得較高
之 Trypsin 和 Chymotrypsin 活性。天然蛋白質水解?抑制物或 Carboxypeptidase A 和 B 之
基質鏡像異構物,可固定於廉價蟹殼幾丁質而製備成各種親和吸著劑。當活化之胰臟粗抽提液連
續通入裝有幾丁質-CHOM,幾丁質-STI,幾丁質- -amino-n-caproyl-L-Tyr-D-Trp 及幾丁質
- -amino-n-caproyl-D-Arg等親和吸著劑之一系列親和層析管柱時,四種蛋白質水解?Trypsin,
Chymotrypsin 及 Carboxypeptidase A 和 B 可被分別吸著和溶離,以微量試驗時,同收率約
80~90%,由此純化出之 Trypsin 和 Chymotrypsin,經圓柱膠體電泳法鑑定時 Chymotrypin
已接近均一純度,Trypsin 有兩個主要環帶,但均具活性,是為α和β Trypsin。若以酸性-
乙醇法或食鹽-乙醇法抽提胰臟胰島素時,蛋白質水解?原亦溶於乙醇溶液而變性,同時由一胰
臟抽提胰島素和蛋白質水解?的方法有待進一步研究。蟹殼幾丁質很適合於倣為親和吸?劑製備
之不溶性擔體材料,惟此種天然聚合物必須保存於潮濕狀態,經脫水處理而保存時,因不完全可
逆之收縮作用而失去其做為不溶性擔體之優良性質,此點可由掃描式電子顯微鏡觀察顯示。
e - 英文摘要: Hog pancreas were prepared in the powderform by an acetone-drying method. Approxi-mately 10 to 15g of pancreatic powder wasobtained from, 100g of fresh hog pancreas. Theproteolytic zymogens of hog pancreas were ac-tivated either in minced tissue or in the extract ofthe acetone-dried powder. When the zymogenswere activated directly in minced tissue, theactivities of carboxypeptidase A and B obtainedwere higher. On the other hand, when thezymogens were activated in the extract of theacetone-dried powder, the activities of trypsinand cbymotrypsin obtained were higher. Severalaffinity adsorbents were prepared by couplingnaturally occurring proteinase inhibitors or enan-tiomers of substrates of carboxypeptidase A andB to an inexpensive water-insolube support chitin.When 1ml of activated hog pancreas extract waspassed successively through a series of columnspacked with affinity resins chitin-CHOM, chitin-STI, chitin-ε-amino-n-caproyl-L-Tyr-D-Trp andchitin-ε-amino-n-caproyl-D-Arg respectively,four proteolytic enzymes, trypsin, chymotypsinand carboxypeptidase A and B were adsorbed onand eluted from each column respectively. Theproduct recovery was about 80 to 90% in thissmall-scale operation. The isolated trypsin re-vealed α and β forms while the chymotrypsinwas almost homogeneous as examined by discgel electrophoresis. If insulin was extracted with 70% acidicethanol solution or salt-ethanol solution,theproteolytic zymogens were also extracted anddenatured. For the best utilization of hogpancreas, further investigation is required toestablish a systematic fractionation of insulinand proteolytic enzymes from hog pancreas. After the preparation, the chitin particlesshould be kept moist; removal of Water by anymeans caused irreversible shrinkage of gel ma-trices and thus resulted in the descrease of carriercapacity as evidenced by an observation with ascanning electron microscope.
