- 中文摘要: Fructooligosaccharides是蔗糖經由β-fructofuranosidase之果糖轉移反應,結合1~3分子的果糖於蔗糖所形成之寡糖類,除了擁有蔗糖之可口甜味和良好的食品加工特性外,並具有優於蔗糖之各種生理特性,是一種新的食品素材。本研究之目的即針對fructooligosaccharides之工業化生產進行探討,第一階段工作主要是進行β-fructofuranosidase生產菌株之分離與篩選,所獲致之成果如下:由自然界中分離得到2,250株野生黴菌和酵母菌,其中能產生多量β-fructofuranosidase之菌株有601株,佔總分離菌株之26.7%,而具有高果糖轉移活性(果糖轉移率超過50%)之菌株有120株,佔總分離菌株之5.3%,且皆屬於黴菌。由此120株菌株中複選得5株優良酵素生產菌株,經菌學鑑定結果分別為Aspergillus japonicus, Penicillium lividum, P. frequentans, P. malacaense和P. vinaceum。此5菌株在高濃度蔗糖溶液(50%,w/v)中反應時,其反應產物痙薄層色層分析法展開後,初步判定其中之主要產物為fructooligosaccharides和葡萄糖。 分離菌株中,以A. japonicus NTU-1249菌株具有最強之酵素活性。在28℃, 125 rpm振盪培養3天後,其果糖轉移活性高達44.7 U/ml,果糖轉移活性/蔗糖分解活性之比值達5.72其中90%之酵素活性是存在於菌體內,只有約10%酵素活性存在於培養液中。以含有菌體之培養液在50% (w/v)蔗糖溶液中反應時,其果糖轉移率高達85.1%。以不同的碳水化合物作為菌體生長之碳源時,A. japonicus NTU-1249只有培養於含有蔗糖或菊糖(inulin)之培養基中才能產生高的果糖轉移活性。
- 英文摘要: From 2,250 strains of fungi and yeast isolated from natural habitats, 601 strains with high β-fructo-furanosidase produetivity were selected. Among them, 120 strains of mould have the ability to produceβ-fructofuranosidase with high transfructosylating activity and can synthesize significant amounts offructooligosaccharides. From this group one Aspergillus and four Penicillia were selected as enzymeproducers. They are identified as Aspergillus japonicus, Penicillium lividum, P. frequentans,P. malacaenseand P. vinceeum. The products from sucrose, produced by the five selected microorganisms, are identifiedas fructooligosaccharides and glucose by thin-layer chromatography. A. japonicus NTU-1249 has been found to be the best producer of β-fructofuranosidase with hightransfructosvlating activitv which can produce 44.7U∕ml of transfructosylating activlty and 7.8U∕ml ofhydrolyzing activity after cultivation at 28℃ for 3 days under shaking at 125 rpm. Its transfructosylatlngactivity∕hydrolyzing activity ratio is as high as 5.72. About 90% of the transfructosylating activityproduced by A. japonicus NTU-1249 is bound with the cell and only 10% of it exists in the culture broth.Notably, the transfructosylation ratio of A. japonicus NTU-1249 in 50%(w∕v)sucrose solution is higherthan 85.1%.
