- 中文摘要: 本研究以組織培養法測定蕓苔各部器官,如苗端、葉片、葉柄及花蕾等再生小植株的能力,以瞭解蕓苔無性繁殖的最佳途徑。 苗端培養於含5mg/l BA及0.1mg/l NAA 的培養基中,最有利於側芽的發育而形成叢生苗。如在培養基中再添加 80mg/l的腺嘌呤,則使叢生苗的生長更好。從叢生苗分離的個別苗體,培養於含0.5?1mg/l NAA及0?0.1mg/l Kinetin的培養基中,可誘導苗體基部形成癒傷組織,由癒傷組織再分化?生不定根而形成小植株。培養成的小植株,先移 入保持適當濕度的輕蛭石中生長約10天,移植於土壤中,則能繼續成長發育並能開花。由此法繁殖而得的植株,型態上與 由種子產生者無異,每一苗端經6~8週可產生9倍多的植株,故苗端?無性繁殖的最佳器官。 葉柄及小花蕾(約4 mm長度)亦能分化成不定芽,前者以含5mg/l BA及0,5mg/l NAA,後者以含10mg/l BA 及1mg/l NAA的培養基最能誘導不定芽的形成。培養基中再添加腺嘌呤對葉柄不定芽的發生有抑制作用,但對花蕾不定芽的發生有促進效果。不定芽大多用葉柄基部或花器基部發生,?有極性生長情形。 葉片組織培養於2, 4-D的培養基中,能形成癒傷組織,癒傷組織再經NAA,BA,Kinentin及腺嘌呤的處理,均可由其 表面或內部形成根原體,最後產生不定根,但不定芽的分化?未達成。
- 英文摘要: The requirement of plant growth hormones for regeneration of plantlets from cultured shoot tips, leaf blades, petioles, and flower buds of rape (Brassica chinensis var. Oleifera) was determined. In the shoot tip culture, addition of 5 mg/1 benzyladenine (BA) and 0.1 mg/1 naphthaleneacetic acid (NAA) to the basal medium (modified MS medium) stimulated development of lateral buds which eventually grew into multiple shoots; additional 80mg/1 adenine further enhanced the development. Combinations of 0.5-1.0 mg/1 NAA and 0-0.1 mg/1 kinetin induced the formation of callus tissues at the base end of the individual shoot separated from the multiple shoot. On longer incubation, roots were developed from such callus tissues. Supplementation of 5 mg/1 BA and 0.5 mg/1 NAA to the basal medium usually favored the regeneration of adventitious buds from petioles. At the rate of 10 mg/1 BA and 1 mg/1 NAA, adventitious buds were also formed on flower bud cultures. The medium containing 1 mg/1 2,4-dichlorophenoxyacetic acid caused callus formation on cultured leaf blade. Various combinations of NAA and kinetin or BA and kinetin initiated root primordia from recultured callus tissues, but shoot differentiation from such culture was not observed. Propagation of rape plants through tissue culture is feasible. Plantlets regenerated from shoot tip cultures were grown to maturity, morphologically, they appeared to be normal.
