- 英文摘要: Using Z-Phe-Leu-OTMB as the model compound, the papain-catalyzed peptide bond formation was studied by varying the parameters including pH, reaction time, and concentrations of substrate and enzyme. Optimal yield of the model compound was obtained when Z-Phe and Leu-OTMB were reacted in 0.2 M Mcllvaine buffer-ethanol (7:5 v/v), pH 6.0, at 38蚓 for 6hrs with substrate concentration of 63 mM and enzyme concentration of 25 mg/ml. Under this condition, highly purified Z-Phe-Leu-OTMB was prepared in 92% yield.
