- 英文摘要: Accumulative evidence has supported the role of iron inthe development of atherosclerosis. To test whetheriron-mediated oxidative stress influences plaque stabili-ty, apoliporotein-E (ApoE)-deficient mice (3 months old)were placed on a chow diet or a low-iron diet for 3months, and the abundance of interstitial collagen andthe expression of the matrix degradation-associated en-zyme, matrix metalloproteinase-9(MMP-9),in vascularlesions were assessed.A low-iron diet appeared toreduce iron deposition while substantially increasing col-lagen content of lesions in mice.Immunostaining dem-onstrated lower expression of MMP-9 in lesions of iron-restricted animals. Likewise, SDS-PAGE zymography re-vealed lower gelatinolytic activities in aortic tissues andsera of the same group of animals. When older ApoE-deficient mice(5 months old)received a low-iron diet for2 months, development of the lesion area was not signif-icantly affected.However, the lesional collagen contentwas much higher in the iron一restricted group of animals,and MMP-9 expression in aortic tissues from the samegroup of mice was significantly lower. Treatment ofmurine J774 macrophages with increasing concentra-tions of ferric ammonium citrate significantly enhancedthe amount of MMP-9 secreted.Together, these dataindicate that decreased vascular iron content followingdietary iron restriction in ApoE-deficient mice leads tolower matrix degradation capacity and increased plaquestability.
