倉鼠腦下腺前葉泌乳激素細胞之微細構造

本論文係報導倉鼠腦下腺前葉細胞之一,吾等暫稱為M細胞(可能是泌乳激素細胞)在正常及進行各種不同實驗狀況下的微細構造。 正常倉鼠的M細胞呈橢圓形或錐形,其胞核多偏離細胞中心。胞質內含有豐富而呈平行排列的扁平槽狀顆粒性內質網及發達之高爾基氏器。其成熟分泌顆粒呈圓形或橢圓形(直徑約350~550nm),而未成熟分泌顆粒,其外形極不規則。分泌顆粒以特殊之醣蛋白染法Periodic acid silver methenamine染之無反應。在雄性倉鼠腦下腺M細胞稀少,在雌性倉鼠則反之。 一般已知在懷孕期之雌鼠及經睪丸酮處理之雄鼠等之腦下腺及鼠之腦下腺異處移植塊,均可分泌大量之泌乳激素。本研究觀察此實驗狀況之倉鼠腦下腺M細胞則均呈高度活化的形態特徵;且經睪丸酮處理,除引起雄性倉鼠腦下腺M細胞肥大外,並可增加細胞數目。另外,倘若移走幼鼠,則造成哺乳期雌性倉鼠腦下腺M細胞內分泌顆粒堆積;此與前人報告謂停止哺乳造成雌鼠腦下腺內泌乳激素含量增加是一致的。根據以上之結果,吾等推論倉鼠腦下腺前葉之M細胞可能是泌乳激素細胞。

The ultrastrueture of presumptiVe mammotrophs, tentatively named M Cells, in the adenohypo-physis of golden hamsters is reported here in detail. M cells in normal hamsters are oval or pyramidal inshape and possess an eccentric nucleus, elaborate arrays of cisternae of the rough endoplasmic reticulumand polymorphic immature secretory granules. The mature secretory granules (350-550 nm in diameter)are round, oval in shape and do not react to the periodic acid-silver methenamine method, which is aglycoprotein stain. M cells occur sparsely in male glands, whereas they occur abundantly in females. The following observations made on pituitaries of hamsters under various physiological or experimen-tal conditions suggest that M cells are probably mammotrophs. M cells displayed an ultrastructure indica-tive of a high synthetic activity in pituitaries that are known to produce a greater amount of prolactin thannormal ones, such as those of late-pregnant females, testosterone propionate (TP)-treated males and hetero-topically transplanted pituitaries. In male hamsters, administration of TP produced an increase in thenumber of M cells, in addition to their hypertrophy. One day after cessation of suckling in lactatinghamsters, an accumulation of mature secretory granules in the M cells was observed. This is in agreementwith the previous findlng that nonsuckling induces prolacting storage in the pituitary.
