本研究以 Vincristine(VCR)及 Cyclophosphamide(CPA)為對照,觀察全身加溫治療
(Whole body hyperthermia, WBH)對於 小鼠淋巴性白血病(P388 lymphocytic leukemia)
之療效,及觀察 Tabernamontanine(TAB)對 WBH 此一治療效果之影響〔全身加溫治療(WBH)
依施行方法不同分為 WBH-1, WBH-2,說明見表一腳註。TAB 為一種?(口?)生物鹼(Indole
alkaloid)具有減輕環境溫度昇高時所加之於動物苦楚之作用〕。試驗結果如下:
(一)單獨使用 WBH-1(36.75±0.25°C/60 min/day/10 days)、單獨使用 TAB(15 mg/kg/
day/10 days),或 WBH-1 與 TAB 同一時間合併使用,對 P388 白血病小鼠均無療效(T/C<
125%)。
(二)WBH-1 與 VCR(0.25 mg/kg/day/10 days)同一時間合併治療(T/C=219%)並不能
使單獨應用 VCR 之療效(T/C=217%)提高。
(三)WBH-1 與 CPA(22.5 mg/kg/day/10 days)同一時間合併治療組之部份小鼠可存活一
年以上。一年治癒率(one year curing rate, OYCR)為35.71%。本組之療效為 T/C>1744.2%
,較單獨應用 CPA 之療效(T/C=271.6%)或單獨應用 WBH-1(T/C=108%)為優,故兩者
相互具有良好之協同作用(Synergistic effect)。e - 英文摘要: The synergistic effect of whole-body hyper-thermia (WBH) on Cyclophosphamide (CPA)and Vincristine (VCR) in the treatment ofP388 lymphocytic leukemia in mice hasbeen studied by using CPA and VCR as posi-tive control agents. In our previous study (1),tabernamontanine (TAB) has been shown tohave protective effect against stress imposed byWBH on mice. This effect is evaluated in anattempt to see whether it is of help to benefitWBH treatment. The results are as follows(see also Table 1): 1. WBH-1 (36.75±0.25°C/60 min/day/10days), TAB (15 mg/kg/day/10 days, ip.), orWBH-1 plus TAB exhibited no effect on P388lymphocytic leukemia in mice. 2. WBH-1 exerted no synergistic effecton VCR (T/C of combined therapy=219%as compared with T/C for VCR=217%). 3. WBH-1 exerted synergistic effect whengiven immediately after CPA (22.5 mg/kg/day/10 days, ip.). This group has OYCR of 35.71%.The effect of this combined treatment, in termsof T/C, is 1744.2%, which is much higher thanthat of using both individual treatments alone(T/C for CPA=271.6%, T/C for WBH-1=108%). 4. WBH-2 (36.75±0.25°C/60 min/day/20days) when given immediately after a reduceddaily dose but with prolonged course of CPA(11.25 mg/kg/day/20 days, ip.) exerted no syner-gistic effect (T/C=193%) as compared withthat by using CPA alone (T/C=198%). Theeffect of CPA of prolonged regimen (11.25 mg/kg/day/20 days, ip.), T/C=198%, is considerablylower than that of higher dose given in ashorter course (22.5 mg/kg/10 days, ip., T/C=271.6%).
