生甘藷經常溫糖化生產酒精之研究

以國內盛產之甘藷,利用液化及糖化酵素在60°C, pH3.5 下行糖化,並將所得糖化液以固定化酵母(Saccharomyces cerevisiae)行酒精醱酵。 糖化率在100-200g/1 的甘藷粉末濃度隨濃度之增加而降低糖化率;甘藷粉末濃度150g/1,酵素濃度(以unit/1 α-amylase 及glucoamylase表示),10,000unit/1,經1天糖化,還原糖的生產力可達98g/1-day。 從CSTR之操作結果得知內部質傳阻力對固定化酵母之酒精醱酵速率有甚大之影響,當固定化酵母之直徑分別為0.35cm 及0.25cm 時,每單位固定化酵母體積的最大反應速率(Vm')分別為2.22, 2.44 mole Glucose/1. hr, Effectiveness Factor(η)之範圍分別為0.28-0.5及0.48-0.7。 選用三段錐型反應器,以固定化酵母將糖化液行酒精醱酵,在0.8hr-1之稀釋率下酒精的生產力可達33.5g/1-hr'-選用轉動真空蒸發器當反應器,在7天之半連續醱酵,真空抽取所得酒精濃度可達12-13(v/v)%,殘留還原糖gt6-8g/1,理論收率為85-90%。

Potentiality in producing large amount ofsweet potato in Taiwan motivated the researchon alcohol production. Saccharification of rawstarch was carried out at moderate temper-ature (60°C) without precooking. Theexperiment was performed with liquifyingand saccharifying enzyme (Gluczyme, AmanoPharmaceutical Co. JAPAN) at pH 3.5.Saccharification was followed by ethanol fer-mentation by immobilized yeast(Saccharomycescerevisiae) using Na-alginate as a carrier. Conversion of saccharification decreasedwith increase of sweet potato powder contentin reaction mixture within the range of 100-200 g/1. At concentration of 150g/1 andenzyme dosage of 5ml/1, the production rateof reducing sugar reached 98g/1-day during1 day. Continuous and semi-continuous ethanolfermentation were carried out by usingthree-stage rhombus-shaped bioreactor androtary vacuum evaporator packed with Na-alginate immobilized yeast, respectively. Theethanol productivity of 33.5 g/1-hr wasachieved at dilution rate of 0.8hr withcontinuous fermentation. The ethanol concen-tration it distillate, the residual reducingsugar concentration and the theoretical yieldwere respectively 12-13(v/v)%, 6-8g/1 and85-90% during semi-continuous fermentationof 7 days.
