- 英文摘要: Repeated treatment with amphetamine (AMPH ), a well-known anorectic agent, into animals could induce an-orexia on day 1 and produce a gradual reversion of foodintake (tolerant anorexia) on the following days. It isunknown whether these feeding changes are related todopamine (DA) and/or noradrenergic neurotransmis-sion. Thus, the present study investigated the subtype ofreceptor mediating AMPH-induced anorexia. Daily foodintake was measured after various drugs were given.Pretreatment with haloperidol, an antagonist of DA re-ceptors, may lead to inhibition of AMPH-induced anorex-ia. However, pretreatment with the -adrenoceptor an-tagonist phentolamine, and the -adrenoceptor antago-nist propranolol, failed to modify the action of AMPH, suggesting the involvement of DA receptors but notadrenoceptors in the action of AMPH-induced anorexia.Furthermore, pretreatment with SCH 23390 at a dose suf-ficient to block D receptors or pimozide at a dose suffi-cient to inhibit D receptors blocked AMPH-induced an-orexia, indicating the involvement of D and D recep-tors. In a study of tolerant anorexia, repeated treatmentwith the D /D agonist apomorphine, but not the D ago-nist SKF 38393 or D agonist quinpirole, induced anAMPH-like tolerant feeding response, providing evi-dence for conjoint action of D and D receptors in theeffect. The present results suggest that both D and D receptors are involved in anorexia and tolerant anorexiainduced by chronic intermittent administration ofAMPH.
