- 中文摘要: (1) 水稻發芽種子在R-2或M&S培養基上,2,4-D含量為2,6,15 ppm均能全部 誘發callus(2)水稻callus細胞以cellulase含量,5%, pH5.6-5.8,370C -F&(60rpm) 4小時後,形成原生質量最多,佔6.92%,僅限於callus分生組織內之細胞,才能 分離出原生質,已成長之細胞雖在高濃度cellulase處理下,亦難於形成原生質。 在形成之原生質中,16.13%含有2個以上細胞核。 水稻幼苗未張開嫩葉或黃化幼苗嫩葉以cellulase 2%,pH5.6-5.8,在300C 下震盪(60rpm)3小時後,嫩葉分離出之細胞幾全部轉變為原生質。剛張開之嫩葉 即在高濃度cellulase處理下,亦不能分離出原生質。形成之原生質中29.62%含有 2個以上之細胞核。(3)多核之原生質證明為spontaneous fusion而來,在細胞經過酵素處理,溶去細胞壁 過程中發生。(4) Callus或嫩葉分離出之原生質懸液(protoplast suspension)經0.275M NaN03 與0.033M PEG6000處理後,觀察各處理與對照原生質內所含細胞核之數目,統計 分析結果,顯示各處理對於增加原生質融合之效果不顯著。
- 英文摘要: 1 .Callus tissues were successfully induced from germinated rice seed grown on either R-2 or M&S medium with 2, 6 and 15 ppm of 2,4- dichlorophenoxy acetic acid.2. About 6.92 of 10-days subcultured rice callus cells were enzymatical- ly converted into protoplasts, of which 16.13% were multinucleate. AI- most all of the mesophyll cells of rolled or etiolated young leaves of rice seedling produced protoplasts, among which 29.62 had two or more nuclei.Most of the multinucleated protoplasts were initiated from spontaneous fusion during the process of isolation.3. Protoplast suspension either treated with 0.275 M sodium nitrate or 0.033 M PEG 6000 (polyethylene glycol)for inducing protoplast fusion were found no statistically difference from the control.
