- 中文摘要: [H]-2-Deoxy-D-Glucose ([H]-2-DG)注入尾動脈內,經中樞神經系統攝取後,其代謝產物[H]-2-deoxyglucose-6-phosphate ([H]-2-DG-6-P) R[H]-2-deoxyglycogen無法再進行更進一步的分解,而滯留於神經組織內。這些積聚的放射性代謝物,可藉著自動放射攝影術進行定性與定量分析,並可研究腦及脊髓等局部組織能量的分布及運用的情形。 [H]-2-DG和[H]-2-DG-6-P在組織切片的準備過程中,易溶解於水溶液而流失,僅少量的[H]-2-deoxyglycogen保存於細胞內。著者應用不灌流及數種固定液灌流的方法,冀以獲得最理想的自動放射攝影影像與細胞最高密度的放射性顆粒。 研究結果,以periodate-lysine-paraformaldehyde (PLP)固定液灌流的效果最好。整個腦部構造及放射影像最清楚,黑質與白質的對比也最顯著,顯示腦部細胞攝取且保存著最大量的[H]-2-DG。於顯微鏡下,可觀察到神經組織覆蓋著最高密度的放射性顆粒;在細胞體內的銀顆粒數目要比神經纖維內的多且密集。流失於各種操作液的放射性量也最少 因為PLP固定液能保存最大量的[H]-2-deoxyglycogen於神經組織內;所以是[H]-2-DG最理想的固定液。
- 英文摘要: [H]-2deoxy-D-glucose ([H]-2-DG) injected into animals intravenously was transported to thecentral nervous system by the glucose transport system. The metabolites [H]-2-deoxyglucose-6-phosphate([H]-2-DG6-P) and [H]-2deoxyglycogen cannot be metabolized further and accumulated within thenervous system. The regional variations in the uptake and metabolism of [H]-2-DG in the central nervous systemcan be used for quantification and qualification of local cerebral and spinal glucose utilization by auto-radiographic techniques. During the histological processing, both [H]-2-DG and [H]-2-DG-6-P werehighly water soluble and free to diffuse, only [H]-2-deoxyglycogen remained. For the precise localizationof these labelled metabolites at the cellular level under the high resolution of microscope, several methodswere applied in this study. The best autoradiographic result was obtained from the intracardiac periodate-lysine-paraformal-dehyde (PLP) fixative perfused animals. The detailed morphological structures of whole brain was clearlydelineated and labelled. The sharp contrast between the gray and white matter showed that a lot of [H]-2-DG was taken up and preserved in the nervous tissue. Under microscope, the density of silver grains over-laid in neurons and glial cells were heavier than that in nerve fibers. The loss of labelled compounds insolutions were minimal. The PLP method was best for [H]-2-DG study because it can preserve maximalamount of labels.
