率較低。
未成熟種子之子葉及芽體經無菌播種可以發芽成苗,其一至二月齡之苗,可作為繁殖之材
料。其頂芽在含有 IAA 1 ppm 之修改 LS 培養基下長根成苗。節芽在含有 5 ppm BA 之同樣
培養基下能長出一或多芽,而在含有 IAA 1 ppm 之同樣培養基下長根。經採取莖芽之實生苗由
基部長出新芽,如此可連續探芽五至七次,而由節芽長成之新芽及發根後之頂芽亦可用同樣地採
芽繁殖,此法由一實生苗可繁殖許多具有同樣遺傳性質之苗體。
經修改之 LS 培養基為:加二倍之鐵化合物,另加 glutamine, arginine 各50 ppm,malt
extract 100 ppm,糖 0.5%,adenine 5 ppm。培養條件為 18~20°C,2000 lux 白色日光燈,
12小時日長。
此等試管苗在滅苗之(石至)石,25°C(白天)15°C(夜間),12小時日長,4000 lux 白色日光
燈,蓋上打孔塑膠布(每 100 打 0.5 cm 直徑之圓孔),並澆以稀釋三倍之 knop 氏培養
液,此法可得 95%之成活率。u - 英文摘要: Sasafras randaiense is a valuable forestdeciduous species in Taiwan. Its lumber is ingreat demand for manufacturing high qualityfurniture. Owing to difficulties of seed germina-tion and rooting of its cuttings, the efforts tore-establish new sasafras through conventionalpropagation methods are futile. The only al-ternative left is the cloning by the modern invitro culture technique. The shoots of five-year-old sasafras seedlingcan be stimulated to grow multiple shoots onmodified Linsmaier and Skoog's medium (MLS)containing 60 mg/ kinetin. Formation of rootsis enhanced by the transfer of those shoots tothe liquid medium containing 5 to 10 mg/indolebutyric acid, supported with filter paperbridge. A terminal two-cm section from one totwo month-old seedling develops into completeplant on MLS medium supplemented with lmg/indolacetic acid. The upper part of axillarybud develops into a new terminal after the topbud is removed. The cutting of the new ter-minal shoot is made repeatedly. Emergence oflower axillary bud is stimulated in vitro on themedium supplemented with 5 mg/ benzylade-nine. The elongated acillary shoots can beused as cuttings to produce complete plants orused for starting a new culture to form moreaxillary bud. It is 95% successful to transfer the rootedplants to the sterilized vermiculite in growthchamber which controls the temperature of 25°C(day) and 15°C (night, 12 hr.), and the lightintensity of 4000 lux with fluorescent lamp.
