- 英文摘要: In order to determine if protein kinase C(PKC)plays asignificant role in the stimulant action of thromboxaneA (TxA )on pulmonary vascular smooth muscle, TxA -induced contractile responses were measured followinginhibition of PKC. Rabbits were sacrificed and segmentsof the main trunk of the pulmonary artery were removedand placed within a temperature-controlled(37℃)organbath. Contractile responses that were evoked by a TxA mimetic(U46,619, 0.5μM)decreased by 27 and 35% fol-lowing treatment with the PKC inhibitors, calphostin C(2μM) and staurosporine(200μM), respectively. Theseresults account forthe effect of the vehicle, DMSO, whichwas also found to have a concentration-dependent inhib-itory effect on the U46,619-induced contractions. Theeffects of DMSO alone was subsequently subtractedfrom the previously measured responses to PKC inhibi-tors that were dissolved in DMSO to obtain effects attri-butable to the PKC inhibitor alone. It can therefore beconcluded that inhibition of PKC results in partial attenu-ation of U46,619-induced responses supporting the hy-pothesis that activation of PKC plays a partial role inTxA -induced contraction of pulmonary arterial smoothmuscle.
