本論文係以鋅錪鐵(Zinc-Iodide-Osmium tetroxide, ZIO)反應,一般電子鏡術及超高壓電子鏡術來研究大花鼠,沙
鼠,及倉鼠之連合下器官(Subcommissural organ, SCO)細胞內高爾基氏器及內質網的微細構造及其立體影像(Stereo-
images)。
大花鼠,沙鼠及倉鼠SCO細胞之高爾基氏器係由許多扁平之囊狀構造物平行排列而形成的。高爾基氏器可用Z10反
應染色,並呈現由許多管狀構造物所形成之複雜網狀結構:此乃由於扁平囊之表面有許多穿孔(fenestrations)所致。在
大花鼠及沙鼠,其SCO細胞之胞質內充滿了許多擴大型內質網及少量之扁平型內質網;而倉鼠SCO細胞內則無擴大型
內質網,僅有扁平型內質網。在本研究中,所有動物之扁平型內質網均可用ZIO反應染色;而大花鼠及倉鼠之擴大型內
質網則染不上。
由以上結果吾人得知:以適當之染色方法來染細胞內之特定胞器,所得之超薄切片及厚切片,再利用一般電子鏡術
及超高壓電子鏡術來研究該胞器之立體結構實乃一極佳之方法。本論文亦證明了在囓齒類動物之SCO細胞的微細構造及
細胞化學特性具有明顯之品系差異(Species variation)。

不用立體鏡(裸眼)的立體影像觀察法:
(1)雙眼注視遠方之固定目標(此時雙眼視軸平行),並放鬆眼部肌肉,然後將立體偶(stereo-pair) 照片放入視野內,將兩
眼注視照片,但不能改變視軸,即兩眼以平行之目光觀察。或:
(2)先蓋住左眼(或右眼),然後以右眼(或左眼)觀察立體偶之左邊(或右邊)照片,左右眼反復練習後,再以左右兩眼交叉
(視軸交叉,俗稱?雞眼),同時觀察左右兩照片。
經上列方法練習後,在視野中可見到三張影像,其中央之影像即為立體之影像。o - 英文摘要: Fine structure and stereo-images of the Golgi apparatus and endoplasmic reticulum (ER) in thesubcommissural organ (SCO) cells were visualized by the application of zinc-iodide osmium tetroxide(ZIO) impregnation, conventional electron microscopy and high voltage electron microscopy (HVEM). The Golgi apparatus in the SCO cells of rats, gerbils and hamsters consisted of flattened sacculesstacked in parallel array. It showed a selective staining toward ZIO mixture and might form a complexnetwork of tubular structures because of the presence of numerous fenestrations in the flattened Golgisaccules. The cytoplasm of the SCO cells in the rat and gerbil was crowded by dilated cisternae of the ERwith a few flattened profiles. In the hamster SCO cells, however, the dilated cisternae of the ER were notobserved. Flattened cisternae of ER in all species studied showed a positivity for ZIO impregnation andformed a complex tubular network, whereas dilated cisternae of the ER in the rats and gerbils did notshow any reactivity. It was thus determined that the observation of thin and thick sections selectivelystained with appropriate reagent for defined cellular organelles under conventional electron microscopyand HVEM offered valuable information about three-dimensional organization of the cell. A definitespecies-specific variation of SCO ultrastructure and cytochemistry was also demonstrated.
