- 英文摘要: The effects of fractionated oxidized low density lipopro-teins(oxidized LDL) on the growth of vascular smoothmuscle cells (VSMC)and their relationship to the forma-tion of lysophosphatidylcholine(lyso-PC)as well as theactivation of protein kinase C(PKC)were studied. VSMCwere isolated from porcine aorta by explant culture. LDLwas isolated from porcine blood by sequential ultracen-trifugation and oxidized LDL was obtained by incubatingLDL with 5μM CUS0 at 37℃ for various lengths of time.Our results showed that LDL oxidized for 12 h and elutedfrom fast protein liquid chromatography at 43 min inhib-ited the growth of VSMC, and that LDL oxidized for Ion-ger than 48 h and eluted at 48 min stimulated the growthof VSMC. The formation of lyso-PC in the oxidized LDLcorrelated well with its stimulatory effect, suggestingthat lyso-PC is responsible for the mitogenic effect of oxi-dized LDL. This stimulatory effect of oxidized LDL wasinhibited by staurosporine, a PKC inhibitor. Treatmentwith oxidized LDL increased the activity of membranePKC, but it decreased that of cytosolic PKC, suggestingthe translocation of PKC from cytosol to the membranein the presence of oxidized LDL. These results suggestedthat the oxidized LDL-stimulated VSMC growth was me-diated by the formation of lyso-PC and the activation ofPKC.
