- 英文摘要: The myocardial protective effects of trilinolein, isolatedfrom the traditional Chinese herb Sanchi (Panax notogin-seng), are thought to be related to its antioxidant activity.However, the intracellular mechanism underlying theprotective effect of trilinolein in the heart remains un-clear. In the present study, we investigated the effect oftrilinolein on norepinephrine(NE)-induced protein syn-thesis in cardiomyocytes. Cultured neonatal rat cardio-myocytes were stimulated with NE,then protein content,[3H】-leucine incorporation,and (β-myosin heavy chain(β-MyHC)promoter activity were examined . The effect oftrilinolein on NE-induced intracellular reactive oxygenspecies(ROS)generation was measured with a redox-sensitive fluorescent dye (2',7'-dichlorofluorescin diace-tate)and extracellular signal-regulated kinase(ERK)phosphorylation by Western blotting.Trilinolein inhibit-ed NE-increased protein synthesis, (β-MyHC promoteractivity, and intracellular ROS.Both trilinolein and theantioxidant, N-acetyl-cysteine, decreased NE- and H202-induced protein synthesis, (β-MyHC promoter activity,and ERK phosphorylation.These data indicate that trili-nolein inhibits NE-induced protein synthesis via attenua-tion of ROS generation in cardiomyocytes.
