- 英文摘要: Human brain glutamate decarboxylase 65(hGAD65) wasfound to exist as full-length and truncated forms whenthe glutathione S-transferase-tagged hGAD65 fusionprotein was subjected to factor Xa cleavage. The trun-cated form is produced by cleavage at arginine 69 basedon N-terminal amino acid sequence analysis, and has amolecular weight of 58 kD. It is resistant to further factorXa cleavage or mild trypsin treatment and is more activeand more stable than the full一length form.Both the full-length and truncated forms of GAD are also observed inbrain preparations in the presence of protease inhibitors.Furthermore, full-length GAD could be converted to thetruncated form by endogenous proteases, suggestingthat the conversion of full-length to truncated GAD me-diated by endogenous protease may represent an impor-tant mechanism in the regulation of GABA biosynthesisin the brain.
