杉葉紫菜果孢子發芽及初芽生長之研究

本實驗研究多種不同組台的環境條件對杉葉紫菜 (Porphyra angusta) 果孢子發芽及初芽生長的影響,結果發現在各種環境條件組合如不同的溫度、光強度、鹽度、pH值及明暗期處理,對果孢子的發芽率均有相當的影響。 果抱子在15°C,20°C和25°C的溫度下,發芽率隨著光強度之增加而增高。若溫度提高至28°C以上,則發芽率隨著光強度之增加而減低,甚至導致果孢子分解消失。對初芽的生長情形也有相同的結果。 培養液的鹽度以30 左右最適合果孢子發芽,增加或降低鹽度都會抑制,但低鹽度對其發芽之影響比高鹽度為小。初芽的長度在20 ,26 ,32 ,39 和 46 五種鹽度之間則沒有差異。 在 pH 7.0 之培養液中,果孢子的發芽最佳,增加或減低 pH 值會降低發芽率;pH 6.0以下之培養液不適合果孢子發芽。pH 7.0, pH 8.0 和 pH 9.0 三種培養液對初芽的生長沒有顯著的影響。

The carpospores of Porphyra angusta wereincubated under conditions of 12:12 photoperiodcombined with different temperatures, light in-tensities, pH and salinities for studying the ger-mination of carpospores and the growth ofgerm tubes. Carpospores germinated at 15°, 20°, 25°Cand in some cases at 28°C. At 15°C through25°C; the number of germinated spores increasedas the temperature and light instensity increased.On the 5th day of incubation, the percentage ofgerminated spores reached its maximum, about95%, under the condition of 25°C, 4800 lux. At28°C the percentage of germinated spores de-creased as the light intensity increased, and at4800 lux, all the carpospores underwent lysiswithin 5 days. However, at 30°C, irrespectiveof light intensity, they underwent lysis quickly. The effects of light intensity and temperatureon the growth of the germ tubes were almostthe same as for the, germination of the carpos-pores. The longest germ tubes were about 87 mlong and were found at the codition of 25°C,4800 lux. When the carpospores were incubated intotal darkness for five days, the percentages ofthe germinated spores were generally low, about10% at 15°, 20° and 25°C, and with no germina-tion occurring at 28°C. Meanwhile, the ma-ximum growth of the germ tubes were about25 m, and was found at the condition of 15°C.But when those carpospores were returned tothe conditions with 12:12 photoperiod, theyrecovered their vitality within 7 days. The number of germinated carpospores rea-ched its maximum at salinity 32% and decreasedgradually as the salinity became higher or lowerthan 32 . However, the lower salinities hadless unfavorable effects on the germination ofcarpospores than the higher ones. The number of germinated spores and thelength of the germ tubes were the highest inthe medium of pH 7, and then decreased in themedia of a lower or higher pH. However, acidicmedium was found more detrimental to thegermination of spores and the growth of germtube than basic ones.
