- 英文摘要: Pharmacological therapies targeting fulminant lung in-flammation in acute lung injury (ALI)need to be im-proved.We evaluated the effect of thalidomide, a chemi-cal modulating both acute and chronic inflammation,onALI induced by intravenous administration of lipopoly-saccharide(LIPS)and zymosan in male Sprague-Dawleyrats. Injection of LPS and zymosan induced significantlung inflammation,as evidenced by increased neutrophilsequestration in lung tissue as well as enhanced nitricoxide metabolite(NOX-)production in the serum andbronchoalveolar lavage《BAL) fluid.Lactate dehydroge-nase(LDH)activity and protein concentration in BALfluid were significantly increased after administration ofLPS and zymosan.Pulmonary microvascular permeabili-ty was determined using the Evans blue retention meth-od,which showed a significant increase in microvascularpermeability after LIPS and zymosan administration,indi-cating the development of ALI.Animals that receivedthalidomide(100 mg/kg)2 h prior to LPS injection hadsignificantly reduced pulmonary NOX- production,pul-monary microvascular permeability, and LDH activityand protein concentration in BAL fluid.We therefore con-clude that thalidomide ameliorates lung inflammationand reduces ALI induced by combined LPS and zymosanadministration in rats.
