鞭毛蟲的凝集。本研究使用20μg/ml 的 Concavalin(ConA), Peanut agglutinin(PNA), Ricinus communis agglutinin 120
(RCA), Soybean agglutinin(SBA), Ulex europens agglutinin(UEA),及 Wheat germ agglutinin(WGA)等六種Lectins來
偵測經體外培養在對數生長期收集並以磷酸緩衝液(PBS)二次清洗的蟲體。包括三個不同株(C,R,T)的Trypanosoma
cruzi(TC), Trypanosoma lewisi(TI), Trypanosoma musculi(TM). 由臺灣蝙蝠分離的一種錐蟲(Tb), Leishmania
donovani(Ld), Leishmania major(Lm), Leishmania tropica(Lt), Leishmania mexicana amazonensis(Lma), Leishmania
tarentolae(Lta)及 Crithidia fasciculata(Cf)等蟲體,結果顯示屬Kinetoplastida目之原蟲體表全含 D-galactose 及 methyl-
α-D-mannopy ranoside 而全不含 L-fucose,除 Lta 外全部含有 N-acetyl-D-galactosamine,另外 Tc(T)與 Tb 體表寡醣
分子完全相同,被 WGA 凝集不被 PNA 凝集,Tc(R)則與 T1, Lm, Lt, Lma, Cf一樣,被 PNA 凝集而不被 WGA 凝集
,Tc(C)與 Ld 可被兩者所凝集,但在 Tm 與 Lta 則兩者均不能凝集。所有鞭毛蟲均可被 RCA 所凝集而且很快的就被
殺死。蟲體若先以0.05%胰蛋白?經37°C, 30分鐘處理,其凝集效價會降低,而在有凝集素專一結合的單醣分子存在時
,其凝集可被完全抑制。n - 英文摘要: Crithidia fasciculata, Leishmania donovani, Leishmania major, Leishmania mexicana amazonensis,Leishmania tropica, Leishmania tarentolae, Trypanosoma sp. from Formosan bats (Tb), Trypanosomalewisi, Trypanosoma musculi, and different strains of Trypanosoma cruzi (Tc) were cultivated at 27°Cin a liquid culture medium. Flagellates harvested from log phase culture were analyzed for their lectinagglutinating characteristics with concanavalin A (Con A), Peanut agglutinin, Ricinus communis agglutinin120, soybean agglutinin (SBA), Ulex europeus agglutinin (UEA) and wheat germ agglutinin (WGA). Resultsindicated that all these flagellates might have D-galactose and methyl-α-D-manopyranoside on their sur-face. The presence of L-Fucose, which complexes specifically with UEA, could not be demonstrated on thesurface of these flagellates. Results from quantitative comparison of surface molecules of Tb and theTulahuen strain of Tc suggested that Tb may have more WGA-binding molecules while Tc may have moreConA-binding molecutes. Pretreatment of the flagellates with 0.05% trypsin at 37°C for 30 minutes causedsome reduction of agglutination titers. Cell agglutination with lectins was completely inhibited or reversedin the presence of the specific lectin-binding monosaccharides.
