- 中文摘要: 一株自氰化物廢水中分離得到之Klebsiella oxytoca菌種可利用氰化鉀、四氰化鎳、銨鹽,及氣體氮作為其生長時之唯一氮源。此菌可在含有4mM氰化鉀之複合培養基內生長,但具有較長的生長遲滯期。當以四氰化鎳為氮源時,此菌可在好氧情況下良好生長,而在厭氧時則生長極差。銨鹽可作為良好氮源,於好氧時生長極佳。以氰化鉀誘導培養之細胞,將其細胞膜分離後測試其NDAH氧化酵素活性,發現具有較大的活性;而未誘導的細胞則活性較低。以吸收光譜分析其細胞膜,顯示此菌具有二種胞色素氧化酵素,即胞色素d與胞色素o。當以1mM之氰化鉀加入培養基內時,胞色素d之含量增加為1.55倍,而胞色素o則增為2.11倍。測試其固氮酵素nitrogenase之活性時發現,此菌可在無氮培養基內以厭氧方式進行固氮作用。以四氰化鎳誘導培養之細胞,無論有氧與否,與有氮與否,均能表現出acetylene還原活性。
- 英文摘要: A Klebsiella oxytoca strain was isolated from cyanide-contaminated water. This isolate used potassiumcyanide, tetracyanonickelate, ammonium, or nitrogen gas as its sole nitrogen source. The organism grewwell in complex media containing 4 mM KCN, but the growth rate exhibited a prolonged lag phase. Goodbacterial growth, under aerobic conditions, was obtained with tetracyanonickelate; however, poor growthoccurred during anaerobic cultivation. Ammonia readily served as a nitrogen source, and it providedabundant growth under aerobic conditions. NADH oxidase activity on sonic membrane preparations isolatedfrom KCN-induced cells exhibited higher activity than did those concomitant preparations obtained fromnon-induced cells. Spectral absorbance analyses of similar cell membrane fractions demonstrated the pre-sence of two cytochrome oxidases: cytochrome d and cytochrome o. Growth in the presence of 1 mMKCN culture media increased the concentration of cytochrome d by 1.55 fold and that of cytochrome oby 2.11 fold. Nitrogenase activity revealed that nitrogen-fixation growth occurred only under anaerobiccondition when nitrogen-free media were used. Acetylene reduction activities were detected in the tetracy-anonickelate-induced cells regardless of whether they were cultivated aerobically or anaerobically, or in thepresence or absence of nitrogen gas.
