合成高比活性之放射性黴菌毒素

本研究以簡單之化學反應使T-2毒素及PR毒素轉變為具有放射性。(一)將HT-2毒素與過量之 H-醋酸酐作用可得放射性之HT-2 diacetate,進-步在微鹼性 之氨液中水解,即可獲得放射性之T-2毒素,放射比活性為25.5 mCi∕mmol。(二)PR毒素經水解作用,產生PR醇,再將PR醇與 C-醋酸酐作用,則可獲得放射性之PR 毒素,放射比活性為1 mCi/mmol。

T-2 toxin and PR toxin were label-ed with tritium and carbon-14, respe-ctively, by simple chemical reactions.(1) Acetylation of HT-2 toxin withexcess (3H)-acetic anhydride gave ra-dioactive HT-2 diacetate. Followed bymild alkaline hydrolysis of HT-2 dia-cetate with methanolic ammonia gaveradioactive T-2 toxin labeled on 4β-acetoxy group. The specific activitywas 25.5 mCi/mmol. (2) Alkaline hydro-lysis of PR toxin gave PR alcohol.Acetylation of the PR alcohol with(1- C)-acetic anhydride gave radioac-tive PR toxin. The specific activity was1 mCi/mmol.
