中藥山豆根之成分對於動物降溫作用之藥理實驗

(fractions)對甦醒大白鼠在不同環境溫度下,體溫調節反應的影響,其中包括新陳代謝速率、血管舒縮活動性(由大白
鼠尾巴及腳掌溫度測知)及直腸溫度變化。
山豆根粗提取液或純化之結晶matrine(主要之生物鹼),經腹腔注射於大白鼠在環境溫度8°C及22°C時,皆有降
溫作用。且降溫作用是由於代謝率降低以及血管擴張所引起。然而在30°C時,?無降溫作用,除此,matrine引起降溫
作用不能被5,6-DHT或6-OHDA所抑制。我們由實驗結果顯示,山豆根之有效降溫成分為生物鹼部份的matrine,並
建議matrine引起降溫作用可能與中樞神經釋放Serotonin或Catecholamines無關連。

In the present study, the 95% ethanolic crude extract of Shan-Dou-Gen was systematically fraction-ated. From it, several components such as β-sitosterol, stigmasterol, lupeol, β-sitosterol-β-D-glucose, 1-maac-kiain, sophoranone, sophocarpine, trifolirhizin and matrine were isolated and identified. The effects of intraperitoneal administration of each fraction, subfraction, or pure compound ob-tained from Shan-Dou-Gen on the thermoregulatory responses (including metabolic rate, vasomotor activityand rectal temperature) were assessed in conscious rats at various ambient temperatures (Ta). It was foundthat i.p. injection of matrine or crude extract of Shan-Dou-Gen produced hypothermia in rats at Ta of8°C and 22°C. The hypothermia in response to matrine was due to a decrease in metabolic rate and/or anincrease in cutaneous temperature. However, i.p. injection of matrine or crude extract of Shan-Dou-Genproduced an insignificant change in thermoregulatory responses in rats at Ta of 30°C. In addition, thehypothermic effect by matrine was not antagonized by pretreatment of rats with 5,6-dihydroxytryptamineor 6-hydroxydopamine. Our data indicate that matrine is the active hypothermic constituent of the root ofSophora subprostrata, and suggest that the hypothermic effect by matrine could not be associated with thecentral release of serotonin or catecholamines.
