- 中文摘要: 在數種系統中,α2-腎上腺接受器之協同劑(agonist)及神經胜Y(NPY)彼此間可相互加強彼此之作用,我們最近曾經報導NPY為一孤立束核(NTS)內之一強效降壓劑,本實驗之目的即在探討NPY與α2 agonist 在NTS內之相互調節作用。實驗中,60 n1之不同劑量之NPY,NPY抗體,α-MNE,Clonidine 及α2對抗劑 idazoxan,yohimbine微量注射到NTS內以觀察其對心臟血管系統之影響。實驗結果發現α2對抗劑idazoxan 及 yohimbine 均可顯著的減低NPY之強效降血壓及心跳之功能。而微量注射NPY抗體則可抑制中樞降血壓藥物,α-MNE及 Clonidine 之降壓效果,但失去活性之抗體則不會有此作用。此結果顯示NPY與α2 agonist間在腦幹有彼此加強之作用,建議NPY與 catecholamine 在中樞心臟血管之管制上有相互作用。
- 英文摘要: In several systems, (-adrenoreceptor agonists and neuropeptide Y (NPY) potentiate one another.We reported recently that NPY is a potent depressor agent in the nucleus tractus solitarii (NTS). Thepurpose of this study is to investigate the possible modulation of the agonist effect by NPY in this site.Microiniection (60n1) of NPY, anti-NPY antiserum, the asonist α-methylnorepinehrine (α-MNE),clonidine, and the antagonists idazoxan and yohimbine were maed into the NTS. Administration ofidazoxan (0.2 nmo1) prior to the injection of NPY (2.3 pmo1) attenuated the potent depressor and brady-cardic effect of NPY. There was a similar attenuation of yohimbine's effect. Similarly, prior administra-tion of the anti-NPY antiserum attenuated the depressor effect of the central antihypertensive agents, α-MNE and clonidine, whereas inactivated antiserum or control normal rabbit serum were not ableto attenuate these effects. Even a subdepressor dose of NPY (47 fmo1) could potentiate the effect of α-MNE.These results demonstrate a reciprocal potentiation of NPY and agonists in the brainstem, andsuggest that NPY and catecholamines interact in central cardiovascular regulation.
