- 中文摘要: 很多起癌性化合物要在細胞中新陳代謝活化之後,才能發揮癌化作用。吾人研究癌化之機轉時宜使用活化型之物質,才能獲得正確的癌化真象。N-Bzo-MAB為合成的酯類,非常的活潑,多方面證明它為起癌色素酪黃素及MAB之類似活化型物質。本研究觀察到N-Bzo-MAB對谿胚之發育有明顯的畸型作用,如產生骨骼的短小,羽毛生長之受阻,腳爪之彎曲以及水腫的現象。MA B雖然對雞胚有毒卻不會產生畸形作用。同時N-Bzo-MAB可引起鵝胚中羽毛及膠原蛋白之減少及自由態翔脯氨酸之增加,這些生化變化與形態上的變化可以互相呼應。因經脯氨酪之增加可能由於膠原蛋白形成受阻所致而膠原蛋白之減少可影響到表皮,骨骼及羽毛之分化,因此組織分化受阻是否可導致癌化,值得進一步的探討。
- 英文摘要: The carcinogen N-benzoyloxy-N-methyl-4-aminoazobezene(N-Bzo-MAB) is an excellent prototype of the metabolized active form of N-methyl-4-aminoazobenzene (MAB)·Single doses of N-Bzo-MAB adminis-tered to the yolk sac of the -chick embryo on the first to 12th day of inc-ubation produce a variety of teratogenic disturbances in the embryo depe-nding on the dosage and the time of injection·MAB is also found to beembryotoxic but does not produce teratogenic symptoms·N-Bzo-MAB isrelatively more toxic and teratogen ic in the early embryos(1st-7th day)and in those that survive,weight inhibition,defects in feather development-flexed claw and edema occur·N-Bzo-MAB cause significant increment offree hydroxyproline in the embryonic tissues. The teratogenic effect of N-Bzo-MAB is thought to be mediated from the disturbance of collagenmetabolism.The association of collagen metabolism with carcinogenesis isdiscussed.It is suggested that suppression of collagen may upset the normalpathway of cellular differentiation and specification and consequently mayinduce the neoplastic change in the affected cell.
