- 英文摘要: We have previously suggested that thymosin αl (thyα1),an immunomodulating thymic hormone, can activatetumor-associated macrophages to a tumoricidal state ina murine model bearing a transplantable T-cell lympho-ma of spontaneous origin designated as Dalton's lym-phoma(DL).Since tumor-infiltrating dendritic cells(DC)also play an important role in the host's antitumorresponse and are as such in an immunocompromisedstate in a tumor-bearing host, in the present investiga-tion we studied if thya1 is able to influence the differenti-ation of tumor-associated macrophages (TAM) into DCwith granulocyte macrophage colony stimulating factor(GM-CSF),interleukin (IL)-4 and tumor necrosis factor(TNF) and whether these TAM-derived DC show en-hanced antitumor activity. It was observed that DC gen-erated from thya1-administered tumor-bearing miceshowed augmented antitumor activity in vitro. Adoptiveimmunotherapy using TAM-derived DC showed a signif-icant delay in the tumor growth and a prolongation of thesurvival time in tumor-bearing mice.DC obtained fromTAM of thya1-administered mice also produced an en-hanced amount of cytokines like IL-1 and TNF-α. This isthe first study of its kind regarding the effect of thya1 onthe differentiation of DC from TAM and the role of TAM-derived DC in tumor progression.
