利用米酒廠廢液進行檸檬酸醱酵

米酒廠蒸餾廢液可被 Aspergillus niger F-0213 用做檸檬酸醱酵之培養基。由於廢液含糖量低,必須添加適當之碳源。本實驗選擇蔗糖做為碳源。磷酸鹽之添加,對菌絲生長量只有少許影響,但檸檬酸之產量降低很多。添加4%甲醇或0.05%黃血鹽,很明顯地提高了檸檬酸產量。微量元素鋅鹽之添加,不但不會提高檸檬酸產量,且與鎂鹽同時存在時,反而有抑制作用。添加氮源,可增加菌絲生長量及糖消耗量,而唯有加入低濃度(0.2%)之硝酸鈉或尿素,能提高檸檬酸產量。廢液之 pH 值若調高至 pH4,則檸檬酸產量達消耗糖之 73.59%。除不添加氮源外,在6%糖濃度下,總合上述有利條件,且接種砲子懸浮液 2.4ml/100m1 之培養液,於28°C表面培養,會使培養液之 BOD 降低 81.55%,產酸達消耗糖之80.29%;對原料糖收率為78.15%。

The distilled slop of rice liquor wasused as a fermentation medium for theproduction of citric acid by Aspergillusniger F-0213. Since the sugar contentin the distilled slop was low, the addi-tion of sucrose as a carbon source forthe fermentation is needed. The addi-tion of phosphate appearedto have little effect on mycelial growth,but markedly reduced the formation ofcitric acid. Methanol at a concentrationof 4% or potassium ferrocyanide at0.05% can increase the amount of citricacid markedly. The element magnesiumcould stimulate the production of citr-ate, however, the addition of zincelement not only had no effect oncitrate formation, but cancelled out thestimulation effect caused by magnesiumwhen elements zinc and magnesiumadded together. The addition of nitro-gen cnmpound increased mycelial grow-th and the consumption of sugar, butonly small amount (0.2%) of orurea added could increase the amountof citrate produced. The pH value offermentation medium adjusted to pH4.0could produce the citric acid at a yieldof 73.59% based on the sugar cons-umed. When the spores were inoculatedat a concentration of 2.4ml/100ml inthe distilled slop with the addition of0.25g/1 , 4% methanol,0.05% Ferrocyanide and 6% sucrose atpH4/0. the yield of citric acid couldreach to 80.29% based on the sugarconsumption and 78.15% on the sugaraddition. After the citric acid fermen-tation, the BOD of distilled slop couldreduce 81.55%.
