本試驗選用水稻為材料,進行原生質體培養基組成分檢討及原生質體細胞壁之再生觀察等研
究。玆將所獲結果摘要?述於後:
本試驗檢討培養基中氮素源與微量生長調節素種類與濃度對水稻原生質體生長之影響。當改
變 MS(Murashige and Skoog)培養基中氮素源時,若完全使用硝酸態氮,有利於原生質體延
長存活;若完全使用銨態氮,原生質體存活數目顯著減少;完全不含氮素,則原生質體消失極迅
速。培養基中必頃添加適當的微量生長調節素(indole-3-acetic acid, naphthalene-l-acetic acid,
2, 4-dichlorophenoxy acetic acid, 6-furfurylaminopurine, -benzylaminopurine, abscisic
acid),可延長原生質體的存活,若不添加任何生長調節素,則於培養初期,原生質體易有出芽
分生現象,但隨之迅速死亡。
水稻原生質體經培養後,雖有細胞壁的再形成,但經 Calcofluor white 染色鑑定,並無螢
光反應,此種細胞壁成分並非以纖維素為主,與組織內細胞壁成分極為不同。
水稻原生質體雖於培養後,有生長現象,並進行數次出芽分生,但無法繼續保持長時期的生
長,仍有待更深入的探討。i - 英文摘要: Studies were conducted to examine theculture performance of rice protoplasts. Theresults obtained were summarized as follows: Nitrogen was an important component forprotoplast growth. By using nitrate as nitrogensource in culture medium, the protoplasts grewwell with active budding and division. By usingammonium salt as nitrogen source, the pro-toplasts grew less and survival protoplasts wereapparently reduced. If nitrogen was withdrawnfrom culture medium, the protoplast could notgrow at all and died sooner. Another important factor of culture mediumwas growth regulator. Without any growthregulator in the medium, the protoplasts showedactive growth and budding during initial 10days, but died very soon. Adding proper amountof growth regulators (IAA, NAA, 2,4-D, kinetin,BA and ABA) to culture medium could prolongthe protoplast survival. Although the protoplastcould be budding and dividing several times,growth and division were discontinued duringprolonged culture, and protoplasts graduallydied out. Because the regenerated cell wallaround the protoplasts could not be detected bycellulose affinity stain, the cell was apparentlydifferent from that of original tissue cells. Thiswas possibily one of the reasons why protoplastsproceeded budding and did not divide normally. The results suggest that the isolated riceprotoplasts are active and viable. The techniquesfor isolation and culture would be extremelyuseful for physiological, biochemical and gene-tical researches of rice protoplasts. However,further studies are needed for successful cultureof rice protoplasts.
