- 英文摘要: Dextromethorphan(DM),an antitussive agent, has beenclaimed to have anti-inflammatory and immunomodula-tory effects in vitro.Thus, the aim of this study was toevaluate the effects of DM on sepsis induced by intrave-nous(i.v.)administration of lipopolysaccharide(LPS)inanesthetized Wistar rats and by intraperitoneal adminis-tration in conscious ICR mice.Results demonstrated thatpretreatment with DM(1,5 and 10 mg/kg,i.v.)significant-1y attenuated the deleterious hemodynamic changes(e.g.,hypotension and tachycardia)in rats treated withLPS.Meanwhile, DM(5 mg/kg)significantly inhibited theelevation of plasma tumor necrosis factor-a and interleu-kin-10 levels,as well as values of GOT and GPT(as anindex of liver function),and BUN and creatinine(as anindex of renal function)caused by LPS.The induction ofinducible NO synthase and the overproduction of NOand superoxide anions by LIPS were also reduced by DM.Moreover, infiltration of neutrophils into the lungs andliver of rats 6 h after treatment with LPS was alsoreduced by DM.In conclusion,the beneficial effects ofDM on LPS-induced sepsis result from its anti-inflamma-tory and antioxidant effects. Thus, DM can possibly beused as a prophylactic agent for sepsis in the future.
