• 中文摘要： 在DNA重組技術日漸發展的趨勢下，以RFLP（restriction fragment length polymorphisms）資料為標識基因，進行遺傳疾病連鎖基因定位以應用於臨床遺傳疾病諮詢，已成為人類遺傳學分析上的一項重要工作。目前國際上所採用的連鎖分析方法通稱為LODS，在分析實際家庭資料時，通常以測得之LODS大於3視為標識基因與性狀基因（如疾病因子）有顯著之連鎖關係。但是以3為臨界值僅為相約成俗，缺乏進一步的學理支持。本文以複雜雙親交配型之二代家庭為模擬資料，進行分析，探討是否LODS的極限分配近似於卡方分配，並就非平衡狀態之連鎖作相關討論。結果顯示當連鎖處於平衡狀態，或標識基因座之相對基因頻度趨近於0.5時，使用3為判定連鎖存在之臨界值時，易有缺失，應當注意。
