- 中文摘要: 細胞生物學在骨科的領域中,正廣泛的被推展。其中,生骨細胞是骨生成作用中,最重要的細胞,它的培養卻因為失去骨間質而變得十分困難。本研究,成功的發展出新的體外培養方式,使得生骨細胞能理想的生長在覆有膠原質的培養皿中。利用以下的方式來評估其正常之生骨細胞功能:(1)在光源及電子顯微鏡下,呈現生骨細胞具有之結構。(2)細胞四周或胞膜交界,可觀察到膠原質之細纖維。(3)由磷酸鈣染色可見明顯的礦物化作用之進行。(4)與對照組之纖維母細胞相比,此細胞對鈣離子之吸收顯著的增加。由此可證明,此類生骨細胞之培養模式,是提供生骨細胞體外培養研究之理想模式。
- 英文摘要: For the improvement of the adult osteoblast culture, the osteoblasts of young adult rabbit endostealfrom long bones were isolated by collagenase digesting separation. 0.1% of type-I collagen precoatedculture flasks were used as substrate for isolated bone cell growth. Morphological examination of culturedcells under a phase-contrast microscope, SEM and TEM observations showea a structure similar to osteoblastin vivo. Histochemical examination of alkaline phosphatase demonstrated 97% purity of cultured osteo-blasts. The presence of calcium deposit activity in cultured cells was demonstrated by Van Kossa stain.High activity of alkaline phosphatase and inorganic pyrophosphatase in cultured osteoblasts as determinedby biochemical analysis. High calcium uptake in cultured osteoblasts was demonstrated by radioisotopelabelled 4sCaCl12. According to these methods, it was indicated that the cells isolated from young rabbitlong bone endosteal were osteoblast-like and still maintained their biological function. Our system forculturing osteoblast-like cells is a successful attempt in growing bone tissue in vitro starting from isolatedbone cells. Therefore, this modified method for bone cell culture on collagen precoated culture flaskscould be used as the experimental model in studies concerning the osteoblasts in vitro.
