動情激素對子宮頸癌細胞株在抗藥性誘發角色之研究中,細胞經不同劑量之二氫基女素(E)或二乙合成青春
素(DES)培養後,吾人可藉MTT實驗偵測到經Adriamycin處理後之細胞存活率,並以流量細胞計數器估算其細胞內Ad-
riamycin之累積量。在相同條件下,多抗藥性基因mdr-1之表現,可藉由此北方浸潤雜交試驗偵測其mdr-1之訊息核糖
核?酸,及利用免疫浸潤法偵測其通透性醣蛋白170之表現。
本實驗結果顯示,動情激素對子宮頸癌細胞株(SiHa及Caski)抗藥性之誘發確實有其效應,當細胞經動情激素
及Adriamycin處理後,其細胞內Adriamycin之累積量會降低同時細胞之抗藥性會增加。由於SiHa細胞之抗藥性主
要乃經由多抗藥性基因被動情激素誘發其表現所造成,故吾人認為在SiHa細胞內多抗藥性基因之調節區中,可能有動
情激素負責要素存在。然而動情激素對Caski細胞抗藥性之誘發機轉卻與SiHa細胞之機轉相異,其主要差別在Caski細
胞內並未測得多抗藥性基因mdr-1之表現。
除上述所得之結果外,本實驗亦可提供吾人以此模式,進一步探討動情激素與其他女性生殖癌抗藥性間之關係。
t - 英文摘要: The role of estrogenic hormones on the induction of drug resistance was studied in cervical cancercell lines, SiHa and Caski. After the cells were inoculated with estradiol (E) or diethylstilbestrol (DES)in various dosages, the cell survival rates with adriamycin treatment were examined by MTT (3-[4,5-dimethyl-thiazole-2-yl]-2,5-diphenyl-tetrazolium-bromide) test and the intracellular accumulation of adriamycin wasevaluated by flow cytometry. In the same condition, the expression of multidrug resistance gene-1 (mdr-Igene) was detected either by Northern blot hybridization for mdr-1 mRNA or by immunoblot for P-glycoprotein170. The data in this study indicated that estrogenic hormones had the capacity to induce drug resistancein cervical carcinoma cell lines. When cells were treated with estrogenic hormones and adriamycin simul-taneously, the intracellular accumulation of adriamycin declined and corresponded with the drug resistance.Since the expression of the mdr-1 gene induced by E or DES results in drug resistance, it is suggested thatthe mdr-1 gene in SiHa cells may contain the estrogenic responsive element (ERE) in its regulatory region.However, the mechanism of drug resistance induced by estrogenic hormones in Caski cells is different fromSiHa cells due to the absence of mdr-1 gene expression. Despite that, this experiment may provide a modelto investigate the relationships between estrogenic hormones and drug resistance in other female genitalcancers.
