以Zymomonas mobilis生產酒精

1.供試之菌株分別為 Zymomonas mobilis, ATCC 29191, ATCC 29192 及 ATCC 10988。 經選出每公升含有葡萄塘100 g, yeast extract 5 g, 1 g, 1 g 及 0.5 g之Rogers 培養基為酒精生產之培養基。於30°C下24小時之培養,其產量分別 為45.84 g/1 (ATCC 29191), 45.68 g/1 (ATCC 29192) 及 44.40g/1 (ATCC 10988)。2.最適初 pH 值供菌體生長及酒精生產者分別為 5.10-7.30(ATCC 29191), 5.78-6.75 (ATCC 29192)及 5.60-7.25(ATCC 10988).3.不同溫度下培養對酒精之產量影響最大,在 30°C 時之產量最高,在 26°C 時之產量約為 30°C 時之 80-85%。超過 30°C 愈高,產量愈低。4.Rogers氏培養基中之採用不同氯源代替,或增加 yeast extract 之加入量,對酒 精之產量並無顯著之影響,但其中之 yeast extract 可以脫脂豆餅及酵母粉代替。5.以 ATCC 29191 參與不同糖分含量之酒精生產試驗,其結果分別為 46.40 g/1(10%葡萄糖 ),61.07 g/1(15%葡萄糖)及 70.53 g/1(20%葡萄糖)。如將 Rogers 氏培養基中之成 分加倍,則在20%葡萄糖時之酒精產量為 77.92 g/1 較僅增加糖分之結果 70.53 g/1 為高。6.於不同葡萄糖濃度下醱酵,菌株 ATCC 29191 之比生長速率,比葡萄糖攝取速率及比酒精生 產速率等均經計算,以10%葡萄糖時之數值為最高。以 ATCC 29191 與 Saccharomycassp. 396 同在10%葡萄糖醱酵時所得反應動力學參數相比較,獲知菌株 ATCC 29191 之比葡萄糖 攝取速率及比酒精生產速率均較酵母高達4一5倍。

1. Three strains of Zymomonas mobilis, ATCC 29191, ATCC 29192 and ATCC 10988, were used for the present study. The Rogers's medium containing glucose, 100 g; yeast extract, 5 g; , 1 g; , 1 g; and , 0‧5 g in a liter was selected for its high ethanol prodnction by the bacteria. The yield were 45. 84 g/1 (ATCC 29191), 45.68 g/1 (ATCC 29192) and 44.40 g/1 (ATCC 10988) respectively after 24 hr fermentation at 30°C.2. The initial pH values suitable for growth and ethanal production were 5.10-7.30 (ATCC 29191), 5.78-6.75(ATCC 29192) and 5.60-7.25(ATCC 10988).3. In studying the effect of temperature on the production of ethanol, it was observed that the yield of ethanol would decrease either below or above 30°C for all the strains tested. The higher the temperature above 30°C, the lower the amount of ethanol produced.4. Increasing the amount of yeast extract or usiug different sources of inorganic nitrogen instead of in the medium would not improve the yield of ethanol, However, all the yeast extract added to the medium could be replaced by soymeal and dried torula yeast.5. Increasing the glucose concentration from 100 g to 200 g per liter in Rogers's medium, the yields of ethanol were 46.40 g/1 (10% glucose), 61.07 g/1 (15% glucose), and 70.53 g/1 (20 % glucose) respectively. However, using double strength of Rogets's medium, the yield was up to 77.92 g/1 as compared to 70.53 g/1 by increasing glucose concentration only.6. A comparison between the kinetic para- meters for fermentation of 100, 150 and 200 g glucose/1 by strain ATCC 29191 is calculated. The specific rates of glucose uptake and ethanol production are maximal for a glucose medium containing 100 g/1 and the values decrese with in- creasing substrate concentration. A com- parison between the kinetic parameters for fermentation of 100 g glucose/1 by strain ATCC 29191 and Saccharomyces sp. 396 is also given. The specific rates of glucose uptake and ethanol production for strain ATCC 29191 are 4-5 times higher than that of Saccharomyces sp. 396.
