- 中文摘要: ?進一?探討水稻原生質體經分離後培養之問題,本試驗主要檢討不同酵素濃度與滲透壓劑濃度對分離過程之水稻原生質體之影響。?分別以細胞計數器計算一定時間內分離所獲原生質體之數目,並利用螢光色素吸取法及TTC法檢定原生質 體之存活百分率。結果顯示,纖維分解酵素濃度從0%至3%範圍內,水稻原生質體分離數目與存活率均隨酵素濃度之提高 而增加。當滲透壓劑濃度分別調整?:氯化鈉濃度從0.9%至2.4% ,氯化鈣濃度從1.8%至4.8% ,己六醇濃度從0.5M至 1.1 M範圍內,水稻原生質體之分離數目與存活率,均隨滲透壓劑濃度之昇高而呈漸增之趨勢,但原生質體之直徑則隨濃度 之增高而趨小,惟原生質體內細胞內含物較多,液胞化程度較不明顯。
- 英文摘要: Having the intention of improving rice protoplast culture, the effects of enzyme and osmo-ticum concentration on rice protoplast isolation were examined. The number of protoplasts was counted by haemacytometer and the viability of protoplasts was determined by using fluorescein stain method and TTC method. When cellulase concentration increased from 0% to 3%, both the number of isolated protoplasts and the percentage of viable protoplasts increased. Sodium chloride (NaCl), calcium chloride (CaCl2) and mannitol were used as osmotica, and the concentrations were adjusted from 0.9% to 2.4%, from 1.8% to 4.8% and from 0.5 M to 1.1 M, respectively. The results obtained showed that with the increase of osmoticum concentration, both the number of isolated protoplasts and the percentage of viable protoplasts also increased. But the protoplasts were smaller in diameter and had denser cellular contents and less vacuola-tion at higher osmoticum concentration than those at lower osmoticum concentration.
