- 中文摘要: 本篇利用?子對抗老鼠白蛋白(albumin)之Fab-peroxidase結合物之免疫電子顯微鏡法來觀察經過秋水仙素或酒精處理過之老鼠肝?細胞之白蛋白分佈狀況。我們發現白蛋白祇出現於明肝細胞(light hepatocytes),而不出現於暗肝細胞(dark hepatocytes)。在肝細胞內白蛋白出現於附?在細網膜及核膜之核糖體(ribosome)、胞質(cytosol)中,或者附?於其他胞器(cellular organelles)上。我們不能在核膜槽(cisterna of nuclear envelope)中、粗面及光面之細網膜槽內(cisternae of rough and smooth endoplasmic reticulum)、高氏基體,或分泌小胞中觀察到白蛋白的出現。這個研究的結果?烈的指出白 蛋白分泌之機轉?:白蛋白合成於固定在核膜及粗面細網膜上之核糖體,然後直接排放入胞質(cytosol)中,以便分泌到細 胞外,而不經過胞內管腔的途徑(cisternae of cytocanalicular pathway)。
- 英文摘要: Ultrastructural localization of albumin in the liver of rats treated with either colchicine or ethanol was studied by a technique using rabbit Fab-peroxidase conjugate anti-rat albumin. Albumin was found only in light hepatocytes and not in dark hepatocytes. It was present in ribosomes attached to nuclear envelope and rough endoplasmic reticulum, free in the cytosol, or adherent to cellular organelles; but was never observed in the cisternae of nuclear envelope, rough and smooth endoplasmic reticulum, Golgi apparatus, or secretory vesicles. The study strongly suggests that albumin is synthesized in bound ribosomes attached to nuclear envelope and rough endoplasmic reticulum, and is discharged into cytosol directly, instead of into the cisternae of cytocanalicular pathway for secretion.
