清醒老鼠之大腸推進運動測定法

經一種簡單的手術後以便測定清醒老鼠的大腸通過時間。這種實驗動物在輸注生理鹽水,注射新斯的格明(0.5mg/kg) 或四甲基樟烷胺(120mg/kg)下證明所測的大腸運動相當可靠。Long-Evans母鼠在苯巴比士鈉麻醉下做右頸靜脈插管 。另一管則由盲腸進入然後導至大腸前端約1.5cm處。兩導管由皮下在背部2肩甲骨間穿出,然後用熱封閉。2天後俟 動物完全恢復即可測定大腸推進運動。大腸通過時間,即自0.1 ml鋇劑灌入大腸(經由盲腸插管)開始至鋇劑到達大腸末 端(用X-光照相來決定)所需之時間。這種方法的優點曾加以討論。

A simple method employing roentgenography for determining colonic transit time in conscious rats was described. Female Long-Evans rats were anesthetized with sodium pentobarbitone. The right jugular vein was cannulated with polyethylene tubing for drug administration or saline infusion. Another polyethylene cannula was implanted into the caecum and its tip was advanced to the beginning of the large intestine for barium injection. Both cannulae were threaded under the skin and exteriorized at the back near the middle of the two scapulae, and heat sealed. The animals were allowed at least two days to recover consciousness, and were used to measure the colonic transit time. The transit time was then determined as the time interval between the barium injection to the caecum cannula and its arrival at the end of the distal colon. In order to ascertain whether the above-mentioned method was satisfactory, the effect of neostigmine and mecamylamine on the colonic propulsive motility was studied. Due to the results obtained by the administration of such drugs, it was concluded that our method of determining colonic motility in the conscious rat was a workable one.
