- 中文摘要: 染色體C染色帶製備的一般方法是將玻片在染色之前先經鹹液及650G之SSC等處理。家蠅之X及Y染色體用C染色帶的技術可察出有真染色質區存在。本實驗中以細胞學及生化之方法追究C染色帶形成之原因。結果可歸m成下述三點。(1) DNA及nonhis-tones在C染色帶之形成過程中扮演重要之角色,而RNA及histones則不重要;(2)異染色質中之DNA及nonhistones,有某些特性使其對於吉氏染料吸收量比真染色質大;(3)C染色帶形成之原因是在處理過程中染色體上某些染色質被移去,而不是染色體上各部份DNA復性速度之差異。
- 英文摘要: Brief alkaline treatment followed by prolonged 650C SSC incubation isthe general procedure for C-band. Euchromatic regions were revealed byC-band procedure in the X and Y chromosomes of the house fly. Themechanisms of C-band production were studied by cytological and bio-chemical approaches. The following tentative conclusions have been madefrom the available data. 1.The DNA and nonhistones play important partsin C-band production, whereas the RNA and histones play unimportantparts. 2. Some built-in properties of DNA and nonhistones in heterochro-matin are responsible for more Giemsa absorption. 3. The differential diges-tion of chromatin rather than differential renaturation of DNA along achromosome is the key point for C-band production.
