- 英文摘要: Ciliary neurotrophic factor(CNTF)is known as an impor-tant factor in the regulation of retinal cell growth. Weused both recombinant CNTF and an adenovirus carry-ing the CNTF gene to regulate retinal photoreceptorexpression in a retinal degenerative animal, Royal Col-lege of Surgeons(RCS)rats. Cells in the outer nuclearlayer of the retinae from recombinant-CNTF-treated, ad-enoviral-CNTF-treated, saline-operated, and contralater-al untreated preparations were examined for those ex-hibiting CNTF photoreceptor protective effects. Cellapoptosis in the outer nuclear layer of the retinae wasalso detected.It was found that CNTF had a potent effecton delaying the photoreceptor degeneration process inRCS rats. Furthermore, adenovirus CNTF gene transferwas proven to be better at rescuing photoreceptors thanthat when using recombinant CNTF, since adenoviralCNTF prolonged the photoreceptor protection effect. Thefunction of the photoreceptors was also examined bytaking electroretinograms of different animals. Adenovi-ral-CNTF-treated eyes showed better retinal functionthan did the contralateral control eyes. This study indi-cates that adenoviral CNTF effectively rescues degener-ating photoreceptors in RCS rats.
