- 英文摘要: Peroxisome proliferator-activated receptor α(PPARα) is aligand-dependent transcription factor that regulates theexpression of genes involved in lipid metabolism andtransport. Ligands/activators of PPARα, like fibrate-typedrugs, may have hypolipidemic effects. To identify foodthat contains activators of PPARα, a transactivation assayemploying a clone of CHO-K1 cells stably transfectedwith a(UAS)4-tk-alkaline phosphatase reporter and achimeric receptor of Gal4-rPPARα LBD was used toscreen ethyl acetate(EA) extracts of a large variety offood materials. It was found that the EA extract of bittergourd (Momordica charantia), a common oriental vege-table, activated PPARα to an extent that was equivalentto or even higher than 10μM Wy-14643, a known ligandof PPARα. This extract also activated PPARγ to a signifi-cant extent which was comparable to 0.5 μM BRL-49653.The activity toward PPARα was mainly in the solublefraction of the organic solvent. The EA extract preparedfrom the whole fruit showed significantly higher activitythan that from seeds or flesh alone.The bitter gourd EAextract was then incorporated into the medium for treat-ment of a peroxisome proliferator-responsive murinehepatoma cell line, H4llEC3, for 72 h.Treated cellsshowed significantly higher activity of acyl CoA oxidaseand higher expressions of mRNA of this enzyme and fat-ty acid-binding protein,indicating that the bitter gourdEA extract was able to act on a natural PPARα signalingpathway in this cell line.It is thus worth further investi-gating the PPAR-associated health benefits of bittergourd.
