i - 英文摘要: A suitable condition for protoplast isolation from Ulva fasciata consisted of an enzyme combinationof 4% cellulase and 2% macerozyme in a 1.2 M sorbitol solution (osmotic pressure 1371.2 mOs/kg), overa 12-14 h exposure period at 24℃ while rotating on a shaker at 60 rpm. A procedure for purifying theseprotoplasts has been developed in which the protoplast-enzyme mixture was centrifuged with 35% (w/v)Ficoll-400 at 200 Xg for 20 minutes to obtain a clear interface of protoplast. Mannitol (0.84 M) in seawater(osmotic pressure 1791.5 mOs/kg) exhibited the least osmotic stress during the incubation of protoplasts.Using FBA Staining (Calcofluor white), we found that about 81% of the surviving protoplasts had newcell walls after seven days of incubation at 24℃ and 166 (方程式無法摘錄). Furthermore, only 21% of proto-plasts retained their ability to divide and develop into a full plant after two weeks of incubation in graduallyreduced osmotic environments.
