分子間及分子內交聯血紅素功能之差異

分子間交聯血紅素及未交聯之正常血紅素比較顯示出,它們有相似之對氧的親和力(P =9.5 mm Hg),合作效應(n= 2.6),正常之波耳效應以及88%之2,3二磷甘油酸效應。相反之,分子內之交聯血紅素則顯示出較高之氧氣親和力(P = 5.2 mm Hg),較低之合作效應 (n=1.6),10% 之波耳效應,以及68%的2.3二磷甘油酸效應。此種交聯血紅素,性質功能之改變可能由於在血紅素次分子之間形成了共價鍵之交聯橋,在氧化及去氧過程中限制了次分子之空間結構的改變所致。

The intermolecular cross-linked hemoglobin showed similar oxygen affinity (P =9.5 mm Hg), thesame cooperativity (n=2.6), normal Bohr effect and 88% of diphosphoglycerate effect as compared withunmodified hemoglobin (P =10.5 mm Hg, n=2.5). In contrast, the intramolecular cross-linked hemoglo-bin exhibited higher affinity for oxygen (P = 5.2 mm Hg), lower cooperativity (n=1.6), 10% of Bohreffect and 68% of diphosphoglycerate effect. These alternations of hemoglobin properties are probablydue to the formation of cross-linkage between subunits within hemoglobin tetramer which restricts theconformational change of subunits upon deoxygenation and oxygenation.
