- 中文摘要: 本研究建立藥物接受器結合評估模式以評估十種有中樞神經作用之中草藥水抽提物。這十種中草藥分別為柴胡(Radix Bupleuri)、川芎(Rhizoma Chuanxiong)、當歸(Radix Angelicae sinensis)、丹參(Radix Salviaemiltiorrhizae)、獨活(Radix Angelicae pubescentis)、黃芩(Radix Scutellariae)、秦艽(Radix Gentianae macro-phyllae)、升麻(Rhizoma Cimicifugae)、酸棗仁(Semen Zizphi spinose)及洋金花(Flos Daturae)。實驗結果顯示這些水抽提物含有可作用在dopamine(D1與D2)、muscarinic acetylcholine(M1)或5-HT(5-HT1A與5-HT2)等接受器及GABAA接受器上benzodiazepine (BDZ)或GABA等結合處之成分。每一中藥水抽提物主要作用之接受器或結合處如下:柴胡:D2, 5-HT1A, GABA;川芎:GABA, 5-HT1A;當歸:GABA, 5-HT1A;丹參:BDZ;獨活:GABA,5-HT1A, D2, D1;黃芩:BDZ,D1,5-HT1A;秦艽:GABA, BDZ,5-HT1A, D2;升麻:5-HT1A;酸棗仁:5-HT1A, 5-HT2,GABA;洋金花:M1, 5-HT1A, 5-HT2。這些結果提供作用在藥物接受器層面之證據,可用來解釋這些中草藥之中樞神經作用。此外,這些結果亦提供某些資訊,可用來導引從這些中草藥中分離及純化與藥物接受器有作用之化合物。
- 英文摘要: In the present study, we established receptor binding assays to evaluate the water extracts of tencentral nervous system (CNS)-active Chinese herbal drugs. These ten herbal drugs are Chaihu (RadixBupleuri), Chuanxiong (Rhizoma Chuanxiong), Danggui (Radix Angelicae sinensis), Danshen (Radix Salviaemiltiorrhizae), Duhuo (Radix Angelicae pubescentis), Hangqin (Radix Scutellariae), Qinjiao (Radix Gentianaemacrophyllae), Shengma (Rhizoma Cimicifugae), Suanzaoren (Semen Zizphi spinose), and Yangjihua (FlosDaturae). The results indicated that these water extracts contained the principles acting on the dopamine(D1 & D2), muscarinice acetylcholine (Ml), or 5-HT (5-HT1A & 5-HT2) receptors, or the benzodiazepineand the -amino-n-butyric acid (GABA) binding sites of GABAA receptors as determined by receptor bindingassays. The receptors or binding sites which predominantly acted by each water extract are listed as follows:Chaihu: D2, 5-HTIA, GABA; Chuanxiong: GABA, 5-HT1A; Danggui:GABA, 5-HT1A; Danshen: BDZ;Duhuo: GABA, 5-HT1A, D2, Dl; Hangqin: BDZ, D1,5-HT1A; Qinjiao:GABA, BDZ, 5-HT1A, D2; Shengma:5-HTIA; Suanzaoren: 5-HT1A, 5-HT2, GABA; Yangjihua: M1, 5-HT1A, 5-HT2. These results providedevidence to explain the CNS effects of these herbal drugs at the receptor level. Furthermore, these resultsprovided information to direct the isolation and purification of receptor-interactive compounds from theseherbal drugs.
