- 中文摘要: 在本研究中, 藉由陰離子交換置換層析, 利用三團聯共聚物為置換齊以分離.beta.-lactoglobulin A和B。在本研究中所使用的置換齊為甲基丙烯酸系列之兩性電解物高分子(Methacrylic polyampholytes): A/sub 8/M/sub 12/B/ sub 12/, A/sub 16/M/sub 12/B/sub 12/, 乙酯(2-(dimethylamino)ethyl methacrylate), M為 2-甲基丙烯酸甲酯(Methyl methacrylate)。經由測定三團聯共聚物之立體因子、特性電荷與鍵結平衡常數, 並探討兩性電解質高分子之結構與溶液性質且由離子交換置換層析之Steric-mass action (SMA)模式可找出系統之沖提線(elution line)與置換線(Displacement line)。本文定性的討論並且成功的證實利用SMA模式預測三團聯共聚物於離子置換層之應用, 也提供此一新一代高分子-三團聯共聚物於置換層析中, 合理之置換劑之選擇與設計。
- 英文摘要: This study explores the feasibility of using triblock copolymer polyampholytes as a displacer to separate .beta.- lactoglobulin A and B isoforms by anion exchange displacement chromatography. The displacers used herein are methacrylic polyampholytes: A/sub 8/M/sub 12/B/sub 12/, A/sub 16/M/sub 12/B/sub 12/ and A/sub 16/M/sub 24/B/sub 24/, where A is methacrylic acid, B is 2-(dimethylamino)ethyl methacrylate, and M is methyl methacrylate. In addition, the steric factor, characterisitic charges and binding equilibrium constant with the ion exchanger gel of the triblock copolymer are measured and discussed with the structure and solution behavior of the polyampholytes. With the Steric-Mass Action (SMA) model of ion-exchange displacement chromatography, the elution and displacement line can be constructed for the system. Moreover, the behavior of elution and displacement line is qualitatively discussed and successfully proven.
