水稻葉肉單細胞的分離及其生理

利用緩和的機械方法,先將水稻葉片打細,再以果膠分解酵素處理不同品種水稻,包括臺中65號及臺南5號,可分離出具有生理活性的水稻葉肉單細胞,這些單離的葉肉細胞,包括有柵狀細胞和梅花形細胞,它們的形狀係由於存在之細胞壁而定。每克水稻葉片所能分離出來的單細胞數目隨著葉齡及總分離時間而差異,在3小時的分離時間中,每克鮮重葉片可以分離得10 ~10 個單細胞。利用臺中65號水稻34天葉齡的葉子,比用同品種,101天葉齡的葉子,不論是3小時或19小時的分離時間,都可以分離出更多的單離葉肉細胞。比較臺中65號與臺南5號可以分離的水稻單離細胞,其產量並無明顯的差異。 利用四唑藍染色法(Tetrazolium blue test)之還原變色反應,以測定單離細胞之生理活性,亦在文中討論。

A new method was developed for isolationof single cells from leaves of Oryza sativa L.Var. Taichung No. 65 and Oryza sativa L. Var.Tainan No. 5. The isolation was made by mildmechanical pre-grinding of the leaf blades,followed by pectinase maceration of the leaffragments. The isolated single cells were pa-lisaded and lobed in shape as examined byphase contrast microscope. The yields of thesingle cell varied with the ages of the leavesused, and the increase was registered with thetime of maceration. The numbers of cellisolated in 3 hrs. maceration time were about105 to 108 per gram fresh leaves. A 34-day-oldleaf tissue yielded more single cells than 101-day one or the same variety. There is nosignificant difference in the yield of single cellbetween varieties as tested. The application of tetrazolium blue test asindication of physiological condition of theisolated single cells was also discussed.
