- 英文摘要: We have examined the human Cyr61 gene and itsexpression in normal fibroblasts. The core promoter,second intron, and 3' untranslated region (UTR) are high-1y conserved between the human and mouse genes.Cyr61 expression was induced slightly slower but moretransiently in human fibroblasts compared to Rat-2 fibro-blasts. These differences may relate to the absence of aserum response element in the human Cyr61 promoter,and the presence of additional AU-rich elements withinthe 3' UTR. Cycloheximide causes accumulation of hu-man Cyr61 RNA in the absence of growth factors, andEGF prevents decay of transcripts in actinomycin-D-treated cells, which suggests that induction by growthfactors may partially involve mRNA stabilization. Wedetect an alternative RNA in serum-stimu∣ated fibro-blasts containing an in-frame deletion within exon 4which disrupts the thrombospondin type 1 repeat. Con-stitutive expression of the full hCyr61 genomic DNA inrodent fibroblasts causes production of multiple proteinspecies, whereas expression of hCyr 4 produces a sin-gle stable protein of the expected size. We also observedmultiple hCyr61 protein species in normal fibroblasts fol-lowing serum stimulation, indicating that Cyr61 may nor-mally be produced as alternative isoforms.
